Hypocotyl explants of radish (Raphanus sativus L. cv F, Taewang) formed calli when cultured on MS medium supplemented with 2.0 mg/L 2, 4-D, 0.05mg/L kinetin, and 0.5 mg/L BAP. Calli were subcultured for four months at intervals of four weeks and then transferred to media containing various concentrations of IAA, BAP, kinetin and zeatin for shoot organogenesis. Shoot primordia were fomed on Ms medium with the combinations of 2 mg/L IAA and Img/L BAP, 21 mg/L IAA and 6-7mg/L kinetin, and 2mg /L IAA and 1-2mg/L zeatin but regenerated normal shoots were produced only upon transfer to medium containing 2mg/L IAA and 7mg/L kinetin. Excised shoots were rooted when cultured on MS basal medium after one week of culture. Regenerants were successfully transplanted to potting soil and grown to maturity in a growth chamber. Histological observations of shoot forming callus revealed that shoot primordia were initiated with the formation of smaller and isodiametric cell files, and that merismetic activity preceded organogenesis.
