Embryogenic suspension-cultured cells derived from leaf base region of the germinated rice (Oryza sativa L. cv. Nakdong) were cultured in LS221 liquid medium with 2 mg/L 2,4-D, 0.2 mg/L kinetin and 0.1 mg/L GA₃for 4 months. After four days of subculture, filtered suspension-cultured microcolonies of 100-250 μm were electroporated with 400 V/cm for 1.2ms. Electroporated cell clusters divided with cell viability of 85% on LS221liquid medium supplemented with 2mg/L 2,4-D, 0.2 mg/L kinetin and 0.1 mg/L GA₃. After three weeks of culture microcalli were formed with frequency of 70% in the dark at 27℃ on MS25 medium. Under fluorescent light of 30 ± 3 μEm-²s-¹these yellow microcalli transferred to N??-22 regeneration medium, formed green spots with frequencies of 24% and frequency of shoots produced from green spot calli was 52%.
