Frequency of germination and contamination from the seeds of 13 rice (O. sativa) cultivars was investigated. Also frequency of callus induction from the hypocotyl explants of seedling was tested on LS medium supplemented with 2.5 mg/L 2,4-D in the dark at 27℃. Hypocotyl explants of selected 4 rice (O. sativa) cultivars were cultured on LS medium supplemented with various concentrations of 2,4-D. The highest frequency of callus formation was obtained when cv. Koshihikari, cv. Nakdong, cv. Ginbuchal and cv. Ricol were cultured on medium supplemented with 2.5 mg/L, 1.0 mg/L, 1.5 mg/L and 2.0 mg/L 2,4-D respectively. Embryogenic calli of pale yellow, globular type were selected and cultured in AA₂ medium supplemented with 2.0 mg/L 2,4-D, 0.2 mg/L kinetin and 0.1 mg/L GA₃ for suspension culture. Initial suspension culture showed large elongated cells and round cells with granular cytoplasm. The fine suspension cell lines were established after 2 months of subculture.
