Previously, the first gene encoding γ-glutamyl transpeptidase (GGT) of the fission yeast was characterized and its expression was found to be regulated by various stressful agents. In this work, a second gene encoding GGT was cloned and expressed from the fission yeast Schizosaccharomyces pombe. The determined sequence of the cloned GGT gene contains 3.040 bp, and encodes the putative 611 amino acid sequence of GGT. The DNA sequence also contains 940 bp upstream and 289 bp downstream regions. The S. pombe cell harboring the recombinant plasmid pPHJ02 showed about 4-fold higher GGT activity in the exponential phase than the cell harboring the vector only, indicating that the cloned GGT gene is functional.
