In this study, we examined the effect of a nitric oxide(NO)-donor sodium nitroprusside(SNP) on the induction of adventitious root formation in hypocotyl cuttings (primary roots removed) of 4-d-old radish seedlings. The treatment of hypocotyl cuttings with 100 μM SNP caused an increase of radish adventitious root growth, as compared with the non-treated control. This enhancement effect of SNP in adventitious rooting was due to the NO because the SNP effect was prevented when the NO-scavenger methylene blue(MB) was added with SNP. In addition, SNP treatment caused an increase of guaiacol pcroxidase(GPX) and syringaldazine peroxidase(SPX) activities in parallel with the induction of rooting in radish hypocotyl cuttings. From experiments using a cycloheximide, it was also proved that the activities of GPX and SPX could be regulated in the translational level of their enzyme protein synthesis.
