Some characteristics of the chloroplast DNA of Korean wild tea plant were studied. The size of Camellis sinensis chloroplast DNA was estimated to be about 134kb by restriction enzyme analysis. Among the restriction endonucleases used. BamHI resulted in more DNA fragments than other enzymes. 28 fragments with the longest fragment being about 20kb. The chloroplast DNA library was constructed by digesting the DNA fragments with BamHI and then cloning into pBR322. The sizes of the inserted DNA fragmens of the 74 clones obtained ranged from 100 to 4.OOObp, and only 3 clones contained DNA fragments larger than 10 kb. Positive clones were selected by hybridization using probes from spinach and rice chloroplast DNA.
