Helicase family is highly conserved among eukaryotes and defines a family of proteins with similarity to has DNA-stimulated ATPase activity. The family containing consensus seven helicase motifs includes proteins from a variety of species with role in cellular processes such as transcriptional regulation , recombination and DNA repair. To identify a new member of eucaryote helicase , two oligonucleotide primers corresponding to stretches of amino acid sequences conserved among eucaryote helicase were designed. After PCR amplification of Tenebrio molitor cDNA , fragment with length 460 bp were subcloned into the pGEM-T vector. The plasmid construct containing the pGEM-T vector and 320 fragment was designated to pPHTC460. Sequence analysis revealed that the amino acid sequences deduced from pPTHC460 was highly homolgous to those in motif 1 and 3 of CHD1 which belong to CHD1 helicase family. The motif 1 and 3 of helicase play an important role in ATPase and DNA dependant helicase function. Results of this study suggested that there is CHD1 helicase family in T. molitor.
