Xenobiotics such as p-pheylenediamine (PPD) , one of hair dye ingredients, have different responses to human body dependant on absorption, distribution, metabolism, excretion, dose and duration of xenobiotics. PPD causes systemic dermatitis and bladder cancer so on. This study is performed to investigate the severe degree of skin toxicity of applied PPD in tungs tate induced xanthine oxidase(XO) deficient model as for the exposed duration. PPD (2.5% PPD in 2% NH40H) were applied to rat skin (25mg/ 16.5crrr’) two or six times every other day in rats fed tungstate diet for 4 days or 12 days and standard diet. There were determined the increased activity of acid phosphatase (ACP) and the increased content of lipid peroxide (LPO) in PPD applied group two times fed tungstate diet for 4 days(PTC) or six times for 12 days(PPTTC) compared with PPD applied group two(PSC) or six times(PPSC) respectively, and The skin tissue was severe as long as the administrated duration was. In the experimental conditions, PTC and PPTTC showed more severe skin damage compared with the PSC and PPSC respectively wheres XO activity was almost not detected in PTC and PPTTC. In addition, there were more decreased rates of the activities of ROS scavenging enzymes and glutathione(GSH) contents in PTC to PTTC compared with PSC to PPSC In conclusion, the increased severity of skin tissue may be due to the excessive reactive oxygen species(ROS) by the long exposed durations of PPD application and tungstate supplemented diet.
