Liriodendron tulipifera grown under a variety of climatic conditions was very valuable for lumber and veneer but the pollination by insects and the number of filled samaras per cone was very low and the frequency was about 10%. To overcome the low pollination the somatic embryogenesis in these trees showed the potential to produce millions of plantlets. To induce somatic embryogenesis the calli were initiated from the immature samaras of adult yellow poplar trees. Embryogenic calli were cluster of small and dense cells termed proembryogenic masses, whereas non-embryogenic calli were friable. For proliferating these proembryogenic masses these masses were effectively subcultured in 1/2 Litvay medium supplemented with 250 mg/L casamino acid, 250 mg/L L-glutamine and sucrose. As for the effects of different osmotica on the maturation media, the highest number of somatic embryos were produced from these masses when cultured on the medium containing 4% sucrose. The highest frequency of somatic embryo production from these masses was achieved on the medium with free ABA. Activated charcoal (5 mg/L) was also effective on the maturation medium with 0.5 mg/L ABA. Upon transfer to germination medium, somatic embryos developed into plantlets successfully on 1/2 Litvay medium with 1.0 mg/L GA3.
