Liriodendron tulipifera (yellow poplar tree) can attain heights 60 m tall and grow under a variety of climatic conditions. This wood is very valuable for lumber and veneer. The tree is pollinated by insects, and the number of filled samaras per cone is very low in natural stands and seems to be about 10%. To overcome the low pollination somatic embryogenesis of this tree has the potential to produce millions of plantlets. The calli were initiated from the immature samaras of adult yellow poplar trees. For proembryogenic mass proliferation from these calli, 1/2 Litvay medium supplemented with 250 ㎎/L casamino acid, 250 ㎎/L L-glutamine and sucrose was effective. As for the effects of different osmotica on maturation media, the highest number of somatic embryos were produced from PEM when cultured on the medium containing 4% sucrose. The highest frequency of somatic embryo production from calli was achieved on the medium with free ABA. 5 mg/L of activated charcoal was effective on the maturation medium with 0.5 ㎎/L ABA. Upon transfer to germination medium, regenerated plantlets were successfully germinated on 1/2 Litvay medium with 1.0 ㎎/L GA₃. Plantlets were successfully transplanted to potting soil and grown to maturity in greenhouse. These efficient plant regeneration systems of yellow poplar tree established in this study, might be applied to mass proliferation, conservation of genetic resource and genetic transformation for molecular breeding.
