Simple Sequence Repeat을 이용한 포도의 종과 품종 내 유전적 다양성 분석

Simple sequence repeat marker system was employed for assessing the genetic diversity of Vitis amurensis , V. coignetiae , V. flexuosa, V. ficifolia, V. vinifera and V. labruscana. A total of 92 Korea vine domestic cultivars belonged to 65 of V. amurensis, 12 of V. coignetiae, 8 of V. flexuosa, 4 of V. ficifolia, 2 of V. labruscana and V. vinifera were genotyped by using 5 simple sequence repeat markers. A total of 45 DNA bands for 92 vine accessions were identified with 5 SSR primers (scu5, scu10, scu11, scu14 and scu15). The DNA bands were amplified with a range of 5-14 bands and an average allele number of 9. SSR-based clustering analysis in 92 vine accessions revealed four major groups with 57.2% genetic similarity. Group Ⅰ includes 49 cultivars in V. amurensis with similarity coefficient of 0.53. Group Ⅱ, Ⅲ and Ⅳ include 12 cultivars in V. flexuosa with 0.13, 15 cultivars in V. amurensis with 0.16, and 16 in V. coignetiae with 0.17, respectively. Results from from these investigations suggest that Genetic relationship was very near between V. ficifolia and V. flexuosa and also near between V. amurensis and those of V. ficifolia and V. flexuosa. But it was far between V. coignetiae and those of V. ficifolia, V. flexuosa and V. amurensis.
