In this study, we examined the effect of a nitric oxide (NO) donor, sodium nitroprusside (SNP), on the inducement of adventitious root formation in cotyledon explants prepared from 4-d-old radish seedlings. The treatment of the cotyledon explants with 50 μM SNP caused an increase in adventitious root induction and growth, as compared with the non-treated control. This enhancive effect of SNP in adventitious root formation was due to NO in that the SNP effect was prevented when the NO-scavenger, methylene blue (MB), had added with SNP. In addition, SNP treatment caused an enhancement of ac acid phosphatase and amylase activities in parallel with the inducement of rooting in radish cotyledon explants. Furthermore, experiments using a cycloheximide has showed that the activities of acid phosphatase and amylase could be regulated by the SNP in the translational level of their enzyme protein synthesis.
