Oral Toxicity and Skin Safety Evaluation of Blueberry(Vaccinium ashei) Leaf Ethanol Extract

This study investigated the single oral dose toxicity and skin safety, such as a primary skin irritation test, eye irritation test and skin sensitization test, of an ethanol extract of blueberry leaf(BBLE) to evaluate its use as a functional ingredient in cosmetics. In a single oral dose toxicity study, no differences were observed between the control and treated groups regarding clinical signs, weight gain, and gross findings. The results indicated that BBLE did not show any toxic effects at 10 mL/kg in mice, and the LD50 of BBLE was found to be higher than 10 mL/kg in this experiment. The results of a primary skin irritation test and eye irritation test in vitro showed that BBLE is included in the non-irritation area. Skin sensitization test results by Guinea Pig Maximization (GPMA) indicated that there was no allergic reaction. These results suggest that the ethanol extract of blueberry leaf in a 1% concentration can be a useful functional ingredient in cosmetics.
