— The effect of diazoxide on melanogenesis was investigated in B16 melanoma cells. Diazoxide stimulated the production of melanin in a dose-dependent manner. Diazoxide increased intracellular Ca 2+ concentration, and BAPTA/AM significantly inhibited the diazoxide-induced melanogenesis. Diazoxide-induced intracellular Ca 2+ increase and melanogenesis was significantly inhibited by EGTA. Taken together, these results suggest that intracellular Ca 2+ influx may mediate the diazoxide-induced melanogenesis, and that diazoxide may be useful in the management of pigmentation disorder.
