Curcumin-induced Apoptotic Cell Death Mediated by ER Stress in Lung Carcinoma Cell Lines

In the present study, we investigated the effect of curcumin, a major well-known compound isolated from rhizomes of Curcuma longa, on the apoptotic cell death and the potential action mechanism of this effect in human lung carcinoma cell lines. The results showed that curcumin inhibited the cell viability in a concentration-dependent manner in both H292 and H358 human lung carcinoma cells. Treatment of curcumin activated caspase-3 evidenced by increased cleaved protein of caspase-3, and subsequently, induced cleavage of PARP. In line with this, Annexin V/PI staining clearly showed that curcumin-induced apoptosis in both cell lines. Interestingly, CCAAT-enhancer-binding protein homologous protein (CHOP), a key molecule involved in endoplasmic recticulum (ER) stress-mediated apoptosis, was significantly increased by curcumin treatment. Collectively, the results suggest that ER stress via CHOP induction might play a significant role in curcumin- induced apoptotic death of human lung carcinoma cells.
