An experimental study on the cytotoxic effect of various non-bone derived bone substitutes in fibroblast

The morbidity associated with autologous bone graft substitutes. These include both bone derived and non-bone derived substitutes, each with advantages and disadvantages. Non-bone derived substitutes include gold, silver, tantalum, vitallium, stainless steel, aluminum, brass, magnesium, zirconium, lead, platinum, titanium, plaster of paris, silicone, teflon, methylmethacrylate, collagen matrix, calcium phosphate ceramic as hydroxyapatite and tri-calcium phosphate, bioactive glass, calcium sulfate. And hard tissue replacement as polymethylmethacrylate (PMMA) and polyhydroxyethyl methacrylate(PHEMA). The incroporation of a substitute is dependent on biological and biomechanical conditions at the graft site. Therefore we studied cytotoxic effect of Calcium carbonate(BIOCORALⓇ Inoteb, France), Calcium sulfate (CAPSETIM, Lifecore, USA), Hydroxyapatite (InterporeⓇ Interpore Co, USA) and Collagen (CollaCoteⓇ Calcitek Inc, USA) on rat 3T3 fibroblast in culture by neutral red(NR) assay, 5-diphenyl dimethyltetrazolium bromide(MTT) assay and sulforhodamine B(SRB) assay. Generally, calcium carbonate and calcium sulfate were represented less cytotoxity than hydroxyapatite and collagen matrix. We considered that the most cytotoxic material were collagen matrix of four experimental non-bone derived bone substitutes and that the more increasing amount of bone substitutes into the well, the more increasing cytotoxity.
