This study undertaken to observe the effects of methotrexate and 5-fluorouracil on the healing of surgical wound in the white rats of Sprague-Dawley strain. A total of sixty-four rats weighing about 200 gm were used through this experiment. Experimental animals were divided into one control group (8 rats) and two experimental groups (56 rats) of MTX group and 5-FU group which were administrated 2.5 ㎎ methotrexate and 200 ㎎ 5-fluorouracil per kilogram body weight with single dose respectively. And the each experimental group was subdivided into seven groups. In group 1, 2, 3, intramuscular injection of MTX and intraperitoneal injection of 5-FU were performed on 3, 2, 1 weeks after incision. In group 4, 5, 6, 7, incision and suture were performed on 1, 2, 3, 4 weeks after administration of anticancer drugs. Thereafter, rats were serially sacrificed in 1st, 3rd, 7th, 14th day after injection of antineoplastic agents (gr. 1, 2, 3) and incision (gr. 4, 5, 6, 7). The specimens were obtained and processed in a routine manner for Hematoxylin-Eosin stain. After microscopic examination, following results were obtained. 1. In group 1, 2, 3, the healing process was not affected by anticancer drug. 2. In group 4, the healing was delayed about 7 days than that of control group. The delayed healing time due to effect of 5-FU was more significant than in the group of methotrexate administration. 3. In group 5, 6, 7, the healing process was not affected by anticancer drug, but proliferation of fibroblasts and collagen laydown were decreased.
