구강암 및 Bleomycin 투여가 Hamster 타액선에 미치는 영향

Effects of oral cancer and bleomycin on the salivary gland in hamster

This study was undertaken to evaluate the effects of bleomycin on the submaxillary and sublingual glands in Hamster with DMBA-induced oral carcinoma. 38 Golden Syrian Hamsters - about 100 gram of body weights - were used. In 2 animals (Group ⅠA), no treatment was carried in order to investigate salivary gland of normal hamster. In 12 animals (Group ⅠB), Normal saline was painted on left buccal pouch three times weekly for 12 weeks. Epidermoid carcinamas were induced in the other 24 animals in the left buccal pouch by paibting three times weekly with 0.5% solution of 9,10-dimethyl-1,2-benzanthracene(DMBA) in heavy mineral oil. Carcinomas were verified by incisional biopsy. At this time, DMBA-treated 24 animals were divided into 2 groups (group ⅡA & ⅡB). In 12 animals (Group ⅡA), Normal saline (5㎖/㎏) was injected intraperitoneally. In the other 12 animals (Group ⅡB), Bleomycin (5㎎/㎏) was injected intraperitoneally. Bleomycin injection was done to Group ⅠB animals with the same method as in Group ⅡB. The Hamsters were sacrificed 1, 2, 3, and 4 weeks after injection of bleomycin or normal saline. The submaxillary and sublingual glands were removed, and processed in a routine manner for H&E stain, Trichrome stain, PAS stain. After microscopic observation, following results were obtained. 1. Buccal pouch cancer exert no significant influence on the salivary glands over the entire experimental periods. 2. In Group ⅡB, as compared with Group ⅠB, there were more infiltration of chronic inflammatory cells and more anisophyll acini, but those showed no significant change with time lapse. 3. Bleomycin-injected group (Group ⅠB and Group ⅡB) showed vascular congestion in interstitial tissue of glands, whereas shame-injected control group (Group ⅡA) does not shown. 4. In PAS stain, there were no significant difference of strain property between DMBA- treatee group (Group ⅡA and Group ⅡB) and normal saline treated group (Group ⅠB), and no significant difference depending on the injection of bleomycin or normal saline. 5. In Trichrome stain, there were no remarkable difference between each groups, but there were some fibrotic change around the acini of submaxillary gland of Group ⅡB, 3-4 weeks after bleomycin injection.
