Bleomycin이 구강암 Hamster의 타액성에 미치는 영향에 관한 실험적 연구

EXPERIMENTAL STUDY ON THE EFFECT OF BLEOMYCIN ON THE SALIVARY GLAND IN THE HAMSTER WITH DMBA-INDUCED ORAL EPIDERMOID CARCINOMA

This study was undertaken to investigate the effects of bleomycin on the parotid gland of the hamster with oral carcinoma. The Golden Syrian Hamsters -7 weeks of age, about 100 gram of body weight - were used. Twenty one hamsters, developed epidermoid carcinoma in the buccal pouch by painting 0.5% DMBA every other day for 12 weeks, served as experimental group, and fourteen hamsters, treated with saline solution with same method, served as control group. Both the eighteen experimental and the twelve control group hamsters were injected intraperitoneally 5mg of bleomycin per kg of body weight with single dose respectively. The hamsters were sacrificed before administration and 1, 3, 7, 14, 21, 28 days after administration of bleomycin. The parotid glands were removed, and processed in a routine manner for H-E stain and PAS stain. After microscopic observations, following results were obtained. 1. The tissue damage of the experimental group before administration of bleomycin may be construed due to application over dose, painting excessive area, inflammation or tisssue degeneration by cancer. 2. The parotid gland of the control group revealed marked blood conjestion 1 day after administration of bleomycin. 3. The parotid gland of the experimental group showed the most destructive feature on the 3rd day, and began to recover from 7 days after administration 4. The tissue damage of the experimental group was more prominent in the acini than in the ducts. 5. There were dilatation of blood vessels and infiltration of lymphocytes. The latter disappeared 7 days after administration, and the former diminished gradually 14 days after administration. But blood vessels were conjested continuously. 6. In PAS reaction, the acini of the control group showed a little irregular positive reaction, but the ducts were within normal limits. 7. In PAS reaction, the acini of the experimental group showed irregular positive reaction and recovered 21 days after administration, but the ducts showed normal appearances 14 days after administration.
