齒科用 acrylic resin에 依한 結締組織性 被膜에 關한 組織化學 및 生化學的 硏究

HISTOCHEMICAL AND BIOCHEMICAL STUDY ON THE CONNECTIVE TISSUE CAPSULES SURROUNDING DENTAL ACRYLIC RESIN IMPLANTS IN RATS

Clinically, acrylic resin has been widely used for subcutaneous prostheses and denture construction. lt was known that capsule was developed around the resin implants. This study was made of the collagen and acid mucopolysaccharide in the connective tissue capsules following a subcutaneous imbedding of acrylic resin plates in Sprague Dawley rats. The materials used were commercial acrylic resin for denture bases. The cured materials imbedded consisted of a plate implant measuring 12mm×12×mm 0.6mm with its round corners. The 80 male rats were divided into 4 groups of 20 rats, and sacrificed. 14th day, 30th day, 60th day and 90th day after implantation. For the histological and histochemical study, the tissue blocks were fixed in 10% neutral buffered formalin and Carnoy s fluid and embedded in paraffin by vacuum pump. The staining methods employed were hematoxylin-cosin stain, van Gieson stain, Gomori s trichrome stain, alcian blue-hematoxylin stain, toluidine blue stain and Gomori s aldehyde fuchsin stain. For the biochemical study, the connective tissue capsules were homogenized, and the pepsin-digested materials of collagen were extracted by the method of Sykes et al. (1976). Then, seperation of the pepsin-digested material was perfomed with sodium-dodecyl-sulfate-acrylamide-gel electrophoresis. The results were as follows: 1. The connective tissue capsules were developed around the acrylic resin implants. 2. In the 14th day and 30th day group, the stain ability for collagen in van Gieson stain and Gomori s trichrome stain was mild, but in the 60th day and 90th day group, it was strong. 3. The reactivity of acid mucopolysaccharide in the fibrous capsule surrouding implant was increased with acrylic resin implantation periord. 4. Collagen and hydroxyproline contents in the capsules were increased with embedding period. But hydroxyproline content in 60th day group was similar with 90th day group and type I collagen was detected from capsule in the sodium-dodecyl-sulfate-acrylamide-gel electrophoresis.
