Tuberculosis is caused by Mycobacterium tuberculosis. Long culture time of the strain limited early diagnosis of the disease. Polymerase chain reaction (PCR) has become one of the most important techniques in molecular biology, and will be more promising in the future. DNA was extracted from patients’ sputa, and the 123 bp DNA fragment on the chromosomal DNA of M.tuberculosis was amplified specifically just within 30 min by PCR. Optimal amplification conditions for rapid detection of the strain from patients’ sputa were established. PCR sensitivity was tested by amplification of the defined genomic DNA in specimens from patient s pleural fluid and lung tissue sections. However, the fragment was not amplified, suggesting that there was not the strain in the specimens.
