무당개구리 (Bombina orientalis)로부터 유도되는 cold-shock protein

In order to recognize the protein biosynthesis induced from the skin of Bombina orientalis under low temperature condition as a cold stress, SD S-PAGE using 10% polyacryamide gel was carried after cold treatment at 4°C. Protein bands induced under cold shock condition has the molecular weight of 88,76, 45, 30 and 20KDa. respectively. And among these 20KDa protein band was remarkable. When it was cultured under cold condition in vitro, cold shock proteins were synthesized in early times as 30min, lhr, 2hrs but relatively long as 2~7hrs in vivo status. To investigate the cold shock protein synthesis in frog skin for a seasonal change, protein patterns in which individuals in summer under cold traetment and in winter under normal and cold treatment were analyzed with SDS-PAGE. This result shows the same protein bands as CSP. The molecular weight of major CSP (CSP20) was 20.65KDa and has a pI value of less than 6.5. For partial purification of CSP20, crude samples were collected and molecular cut-offed with centriprep. and finally chromatographed with the resins for ion exchange and gel filtration.
