In order to monitor the regulation of metamorphosis related genes, the present study attempted RT-PCR using cDNAs from flounder (Paralichthys olivaceus) larvae and juveniles of the early, mid, late, and complete stages on the process of metamorphosis. A positive control EF-la showed strong expression signal in the mid, late, and complete stages. Type I procolalgen al, a2, and a3 chains, consisting of body structure, strongly expressed in the mid and late stages. BMPl showed an signal in the early and late stages of metamorphosis, the strong expressions of MMP2 and MMP13 were observed in the early, mid, and late stages of metamorphosis, and MMP17 showed a weak expression in the early stage of metamorphosis. TIMP2, which inhibited MMP enzyme activity, showed a similar expression pattern with MMP2. Fibronectin was expressed in the all stages, and β-catenin interacting protein was expressed in the early stage of metamorphosis. Moreover, RT-PCR was tried using cDNAs from various tissues and whole body (9.5cm, 4.9g) to figure out tissue specific expression pattern. A positive control, EF-la was observed in all tissues but relatively weak expression in muscle, liver, intestine, gill, and the whole body. Type I procollagen al and a2 chains were observed in all tissues except for intestine. Type I procollagen a3 chain was found in skin, fin, and eyeball. BMP2 was observed in fin, rectum, and eyeballs. The expression of MMP2, MMP9, and TIMP2 was observed in various tissues. The expression pattern of MMP2 and TIMP2 was same. MMP13 and MMP17 expressed in fin and rectum. Fibronectin was expressed in all organs, and β-catenin interacting protein was expressed in brain, fin, rectum, and eyeball.
