Naltrexone과 Risperidone의 병합 투여가 C57BL/6형 생쥐의 알코올 섭취량에 미치는 영향

Effect of Combined Administration of Naltrexone and Risperidone on Alcohol Intake in C57BL/6 Mice

Objectives:In preventing relapse in alcohol dependence, previous studies suggested that combined administration of two or more drugs which are acting on different neuronal system is more effective than the administration of each drug alone. In order to investigate the effectiveness of combined administration of naltrexone and risperidone in comparison with naltrexone or risperidone alone, this study was carried out by comparing the amount of alcohol intake in C57BL/6 mice co-administered with naltrexone and risperidone with that in C57BL/6 mice with naltrexone or risperidone alone. Methods:In 32 C57BL/6 mice in the state of alcohol dependence, naltrexone 1.0 mg/kg or risperidone 0.005 mg/kg administrated respectively or naltrexone 1.0 mg/kg and risperidone 0.005 mg/kg administrated simultaneously for eight days. The amounts of alcohol intake for 2 hours, water intake for 22 hours, and food intake for 24 hours were measured. Results:1) A significant reduction of alcohol intake for 2 hours was observed when the mice were treated with naltrexone 1.0 mg/kg alone and naltrexone 1.0 mg/kg and risperidone 0.005 mg/kg simultaneously. This effect was significant since the 2nd day and remained significant up to the eighth day of drug administration. When naltrexone 1.0 mg/kg and risperidone 0.005 mg/kg were administrated simultaneously, compared with naltrexone 1.0 mg/kg alone, the alcohol intake for 2 hours had a tendency to be reduced on the eighth day of drug administration. 2) A significant reduction of alcohol intake for 2 hours was observed when the mice were treated with risperidone 0.005 mg/kg. This effect was significant only on the 2nd day of drug administration. 3) No significant different was revealed between the effect of naltrexone 1.0 mg/kg or risperidone 0.005 mg/kg alone and that of naltrexone 1.0 mg/kg with risperidone 0.005 mg/kg on the amounts of water intake for 22 hours. 4) Only on the 2nd day of drug administration, a significant reduction of food intake for 24 hours was observed when the mice were treated with risperidone 0.005 mg/kg. Conclusion:From these results, it is suggested that the effect of combined treatment with naltrexone and risperidone might be superior to that of naltrexone alone in preventing of relapse in alcohol dependence.
