혈흔 및 정액흔 검사를 위한 Peroxtesmo？？ KM 및 Phosphatesmo？？ KM 페이퍼의 유효성 검토

Validation Study of Peroxtesmo？？ KM and Phosphatesmo？？ KM Papers for Identification of Blood and Semen Stains

본 연구에서는 혈흔 및 정액흔 예비검사를 위한 Peroxtesmo？？ KM 페이퍼(이하, Peroxtesmo KM) 및 Phosphatesmo？？ KM 페이퍼(이하, Phosphatesmo KM)의 민감도, 특이성, DNA에 미치는 영향을 기존의 검사방법들과 비교하였다. Peroxtesmo KM은 혈흔에 대하여 LMG 및 Leuco-malachite DISCHAPS™ 시약과 동등한 민감도를 보였으며 Phosphatesmo KM은 정액흔에는 SM시약보다 동등 이상의 민감도를 보였다. 다양한 시료를 이용한 특이성 시험결과, Peroxtesmo KM만 채소즙에 위양성 반응을 보였고, 혈흔 및 정액흔 예비검사 시약들이 DNA에 미치는 영향을 알아보기 위해 qPCR 방법을 이용하여 정량한 결과, Peroxtesmo KM 및 Phosphatesmo KM이 가장 높은 DNA 회수율을 보였다. Peroxtesmo KM 및 Phosphatesmo KM은 사용이 편리하여 범죄현장은 물론이고 법과학 실험실에서 혈흔 및 정액흔을 식별할 수 있을 것으로 사료되었다.

In this study, the authors compared new presumptive tests for blood and semen stains, Peroxtesmo？？ KM paper (Peroxtesmo KM)and Phosphatesmo？？ KM paper (Phosphatesmo KM) with conventional methods for sensitivity, specificity and effect on DNA. Peroxtesmo KM showed similar sensitivity to the LMG and Leucomalachite DISCHAPS™ reagents for the blood, and Phosphatesmo KM showed higher sensitivity than SM solution for the semen. In specificity study, Peroxtesmo KM only showed a false positive reaction in the mixed vegetable juice. In addition, we carried out qPCR to confirm the effect on DNA. Peroxtesmo KM and Phosphatesmo KM paper had less effect on DNA than comparative presumptive test methods. The presumptive test papers for blood stain and semen stain were considered easy to use at crime scene regardless of forensic laboratories.
