Cloning, Expression and Characterization of 2-Deoxyribose Phosphate Aldolase Gene(deoC) from Bacillus sp.168

A deoC gene encoding 2-Deoxyribose phosphate aldolase(DERA) was cloned from Bacillus subtilis 168 into an expression vector and protein was expressed in Escherichia coli. A molecular weight of the expressed recombinant DRRA was approximately 24.5kDa. The recombinant DRRA was purified using Ni-NTF affinity chromatography and then characterized. The degree of purification yield of the purified recombinant DERA were 1.4-fold and 74%, respectively, compared with that of the crude extract. The optimum temperature and pH of the enzyme were 20∼50℃ and pH6.0, respectively. The thermostability of the enzyme showed that more than 42% activity remained after at 40℃ for 24h, but no activity over 60℃ at the same condition.
