The study on enzymatic characterization of peroxidase(POD) produced by recombinant S. cerevisiae GPD-POD1 was carried out. Purification procedures included ammonium sulfate precipitation and DEAE-cellulose column chromatography. Final POD activity and specific activity were 85.0 U/mL and 134.7 U/mg protein. The molecular weight of recombinant POD was estimated to be 53 kDa by SDS-PAGE and native-PAGE. The optimal pH and temperature for the POD activity were found to be pH 8 and 30℃, respectively. The enzymatic properties of the recombinanr POD were examined on isoelectric point, glycoprotein, metal ions and substrate specifiities.
