Sequential study on the first appearance of PrPCWD in TgElk mice model

Chronic wasting disease (CWD) was diagnosed in the Republic of Korea in 2001, 2004, 2005 and 2010. Previously, the incubation period of the pool of two elk CWD cases in 2001 (E190Y+229Y) was estimated as 150±12 days postinoculation (dpi) using transgenic mice over-expressing elk prion protein. In this study, end point titration gave a titer of 105.6 ic LD 50/gram of elk brain. Furthermore, TgElk mice were sequentially culled up to 150 dpi. Surprisingly, PrPCWD appeared first in the brain at 60~75 dpi. This result can be used in designing future TgElk mice studies using inoculums with the similar titer.
