This study was performed to evaluate the difference of immunity obtained by the inoculation routes of classical swine fever virus (CSFV) vaccine (LOM-850). Two month-old piglets, which were antibody negative or had 1:2 to 1:4 maternal antibodies to CSFV, were intraocularly inoculated once with the vaccine virus strain LOM-850 (106.0TCID50/ml) and challenged at 2 weeks later with the virulent ALD strain. The piglets showed clinical and histopathological signs of CSF and also did not obtain immunity. However the antibody levels of protective immunity were obtained when the other piglets were vaccinated two times at 2 weeks interval via intraocular route. But the piglets with 1:32~1:128 maternal antibody titers were not fully boosted to the levels of the protective immunity to CSFV after 2 intraocular inoculations because of the interference of maternal antibodies. To compare the level of immunity to intranasal with intraocular vaccination route, two month-old piglets were intranasally inoculated once with 0.65 ml of LOM-850(106.0TCID50/ml). After two weeks of vaccination, antibody levels rose from 1:16 to 1:256 and the protective immunity to highly pathogenic ALD strain (105.5TCID50/ml) was developed in the piglets. In conclusion, intranasal inoculation route was better than intraocular route to induce the protective immunity of LOM-850 vaccine to CSFV in piglets.
