Toxicity Assessment of UVB using Embryo of Zebrafish(Danio rerio) as an Alternative Animal Model

In this study, the effect of UVB on zebrafish generation was evaluated. Ultraviolet light promotes skin aging by inducing the keratinization of the skin and denaturation of the intradermal collagen and elastin. When severe, it can also be a cause of skin cancer by damaging the DNA of the cells. Zebrafish have 70% resemblance with human genes, as well as the homology between genes and protein is very high. The development process of neural system and various organs resembles with those of humans, therefore zebrafish is actively studied as an animal model for human diseases. In this study, zebrafish embryo was used to test the toxicity on embryo against UV radiation. To test the toxicity caused by UV in accordance of time, UVB (50mJ/cm²) has been set as standard to observe embryo coagulation rate, hatching rate and cardiotoxicity in 60, 210, 310, 340, 370 minutes at 0, 10, 30, 40, 60%. As a result, LD50 of zebrafish embryo on UVB(50mJ/cm²) was shown after having been treated for six hours. By these results, we expect to objectify in vivo toxicity by UV radiation as a means to scientifically evaluate the safety and efficacy of cosmetic and raw materials that require testing for UV radiation toxicity.
