Uptake and Elution Studies of Monofunctional Alkylating Agents into Various Tissues of the Rice Seeds

本 實驗은 EMS, MMS와 같은 效率的인 alkylating agent의 吸收率과 水洗에 依한 浸出程度를 決定하고저 水稻種子에 ¹⁴C으로 表識된 MMS를 處理하여 種子의 各 組織部位에서 調査 硏究된바 얻어진 結果는 다음과 같다. 胚가 發芽를 한 경우는 MMS의 浸透가 擴散에 依하고 前 浸漬한 種子는 mutagen을 容易하게 吸收한다. 한편 正租와 玄米에서 mutagen 吸收差異는 없었다. mutagen 飽和는 30分 乃至 1時間 사이에 나타나고, 處理直後에 切取한 胚에서 飽和程度가 完全種子 處理한 것과 同樣이었다. 水稻에서 突然變異誘起를 爲하여는 種子를 催芽始初까지만 浸漬하는 것이 效果的이다. 生育時期에 吸收率과 感受性이 增加되고 低濃度 mutagen이 좋은 結果를 보인다. 이들 種子 處理末期에는 水洗에 依하여 固定시켜야 하고 이들 種子를 貯藏하려면 0℃ 보다 약간 높은 溫度에서 突然變異率이 높다.
The uptake of MMS into various tissues were studied. At the time, when embryos start to germinate the penetration of MMS is determined by diffusion. Presoaking seeds had easily uptake MMS while no essential difference between hulled and dehulled seeds was found. The mutagen saturation was appeared within 1/2-1 hour while in embryos which were dissected immediately after the treatment, the saturation was equal to whole treated seeds. For the practice of mutation induction on rice by EMS, it is recommended to presoak seeds in water until the beginning of sprouting. At this stage the absorption rate and the sensitivity will be increased and then low mutagen concentrations give good results. At the end of the treatment, the seed has to be fixed by water as post-washing. In case of storage at low temperature just above 0℃, it seems to increase the mutagen efficiency.
