Fusion of Protoplasts Isolated from Mesophyll Cells of N. rustica

Mesophyll tissues of N. rustica were treated with macerozyme and cellulase in 0.8 M Dmannitol, and the isolated protoplasts were centrifused repeatedly to induce protoplasmic fusions. Maceration of tissues and isolation of protoplasts proceeded satisfactorily. Immediately after the centrifugation, multilobed complex protoplasmic bodies and giant round protoplasts were observed. Most of the fusion of protoplasts during centrifugation seemed to occur instantly, but the fusion taking place in later stages took more time to get completely fused. Giant round protoplasts had either a few nuclei or single nucleus of large size.
