As a means of selecting out normal looking opaque-2 kernels from a segregating generation of modified opaque-2 crosses, protein bodies in endosperm cells and in aleurone layers were microscopically observed. Determination of protein bodies in the endosperm cells of normal maize, which had previously been done by sectioning kernels and dissolving starch granules using amylase, were possible without using amylase. Endosperm cells of opaque-2 maize completely lacked protein bodies were distinctly observable in both endosperm and aleurone layer cells and varied in quontity depending on the position of kernels and the location of aleurone layers. The kernels observed could be used for propagation after observation.
