Utilization of Corn as Laboratory Materials in the Genetic Classes

Laboratory materials for use in the genetic class are required to help students understand genetic principles, especially Mendel’s laws. Very few materials are presently used in the genetic classes in Korea. Therefore, the objectives of this study was to develop new genetic materials by using kernel color and shape of corn for the purpose of class use. The results obtained are summarized as follows: 1. The gene for purple kernel was complete dominance over the gene for white kernel. The F₁ between purple and white corn was all purple demonstrating the Mendel’s law of dominance. 2. The gene for round kernel was complete dominance over the gene for sugary gene. The F₁ between round and wrinkled corn was all round, demonstrating the Mendel’s law of dominance. 3. The segregating ratio in the F₂ or testcross was all agreed with the expected ratio and was enough to show Mendel’s law of segregation. That is, 3 to 1 ratio in F₂ and 1 to 1 ratio in the testcross of monohybrid and 9 : 3 : 3 : 1 in F₂ and 1 : 1 : 1 : 1 ratio in the testcross of dihybrid were obtained, respectively. 4. The color of leaves was also segregated as expected in the ratio of 3 green to 1 albino in F₂, showing Mendel’s segregation ratio. 5. The phenotype and genotype of segregates in F₂ or testcross were all like the parental phenotypes and genotypes, demonstrating the Mendel’s law of purity. 6. The phenotypes and genotypes of segregates from the testcross of dihybrid demonstrated that the genes concerned were all acted independently (law of independence). 7. The materials used in the study were all like the pea which Mendel had used, since the genes used in the study were non-alleles and independently acted. 8. The identification of each phenotype was very easy and the corn was convenient to handle. 9. The materials developed here are assumed to be used also for the lab exercise of Chi-square test. 10. It was certain that the materials used in the study can be produced in large amount and used for genetic classes.
