Effect of Gamma Irradiation and Ethyl Methane Sulphonate in Tobacco (Nicotiana tabacum L.)

Seeds of 2 tobacco varieties were irradiated with 6 dosage levels of gamma-ray rang ing, from 5 KR to 30 KR, in an attempt to induce mutations. A chemical mutagen, Ethyl Methane Sulphonate, was also used in this experiment:3 varieties, 4 levels of concentration from 0.5 to 4%, and 2 different length of treatment, 3 and 12 hours. At the dosage level of 25-30 KR, 50 % of plants died 4 weeks after germination, and those survived plants showed considerable seedling retardation of more than 50%. Sensitivity to irradiation was more profound in B.Y 104 than Hicks variety at the seedling stage. Overall reduction of plant height in the field was 10-15% compared to untreated plants. The treatment of 4% EMS for 3 hours reduced survival rate to 50 % and 2% for 3 hours caused 50% reduction in leaf length. Three varieties responded same to the various treatments, but the survival rate and leaf length decreased as the duration and concentration of EMS treatment increased.
