The objective of this study was to identify and compare the genetic variations in natural populations of Pinus densiflora in three islands in Korea. Using starch gel electrophoresis, number and frequency of isoperoxidase bands in mature needle leaves were observed in three subpopulations each from three islands, Jeju, Hongdo, and Ulreung. A total of 11 different bands were observed. Among the three islands, Jeju populations showed highest number of bands(7.8 bands per sample) in average, and 6.2 and 6.1 bands per sample were obtained from Ulreung and Hongdo, respectively. The bands, G(Rf 0,3) and Y (Rf 0.65) were observed in all the samples with no variation. The S band showed the largest variation in frequency which ranged from 10 to 66 %. Jeju populations were considered to be the best natural stands among the populations studied and are recommended to be preserved for gene conservation, judged by their good growth, straight stem, and large genetic variation.
