Fusion of Protoplasts from Two Different Varieties in Rice

These works were carried out to isolate protoplasts and to induce protoplasmic fusion with rice callus tissues derived from uninucleate microspores by anther culture of different cultivars or lines. The haploidal calli were practically induced from 0.38% anthers cultured and the callus induction percentage were variably with cultivars or lines. Callus tissues were macerated well and degradation of cell walls also proceeded satisfactorily, but the solution mixed with 0.56M sucrose, 5% pectinase, and 10% cellulase was more effective than that with 0.5% macerozyme, 0.8M Dmannitol, 0.3% potassium dextrum sulfate, and 10% cellulase. When the protoplasts were transferred to sodium nitrate than to incubation medium, the fusions took place letter. The isolations of protoplasts were much alike, but the protoplasmic fusions were different with cultivars or lines. The protoplasts fused which were cultured on incubation medium for 24 hours were seemed to be regenerated the cell walls.
