Cell Culture of Poinsettia (Euphorbia pulcherrima)

For the induced mutation using cultured cells of poinsettia, effective nutrition medium for cell culture, cell survival and proliferation, and radiosensitivity were investigated. Hormone concentration of CPA 0.2 and BAP 0.2 ppm in MS basal medium was effective for callus formation and single cell production, showing cell survival rate of 74% at 15 days after culture. Cell proliferation in batch culture showed logarithmic growth phase from 5 to 14 days and stationary phase in 15-18 days. LD50% of microcluster by radiation treatment was 30 GY. Meristematic single cells originated from microcluster had higher viability and it was easy to regenerate whole plant for practical utilization.
