Studies on direct induction of pollen plants by one-step culture method in rice anther culture

The direct induction of pollen plants by one-step culture method was evaluated in rice anther culture. In order to increase the rate of green plant induction by the method, effects of cold pretreatment and compositiop of auxin and kinetin in media were studied. Cold pretreatment at 4℃ was more effective than 8℃ or 12℃, but the effect of plant hormones was not clear. N6 medium supplemented with NAA 1mg/l, 2,4-D 0.02mg/l and kinetin 0.1mg/l had the highest induction effect as compared to the other combinations of auxin and kinetin, and the rate of green plant induction was 5.2%, on the basis of anther number, using rice cultivar Tatsumi mochi. The efficiency and potential of one-step culture method were discussed.
