Selection in vitro for herbicide-tolerant cell lines of sesamum indicum Ⅰ. Effects of explants and hormonal combination on callus induction

This experiment was carried out to find proper explants and plant growth regulators for profuse callus induction in aiming to select in vitro for herbicide-tolerant cell lines of sesame. Hypocotyl and roots of seedlings were proper explants and shoot tip of seedlings or seed could be also used as explants. There was not much difference in callus induction between 1mg/l and 2mg/l of NAA, and combined effects with BA(0.25-0.5mg/l) was not great. No difference was observed in callus induction between 1mg/l and 2mg/l of 2, 4-D, and callus induction was not much improved when it was combined with kinetin(0.25-0.5mg/l).
