Studies on anther culture of oryza sativa L. Ⅴ. Increase of anther culture efficiency by physiological stress of donor plant in Tongil type cultivars

For the increase of anther culture efficiency, the environmental conditions for donor plants on the androgenic response and on the pollen dimorphism in vivo was investigated in three tongil type cultivars Seogwangbyeo, Samgangbyeo, and Pungsabyeo, which had low sporophytic potential. Donor plants were grown under two day-lengths, 8 and 16 hours and three nitrogen levels, 0, 15, and 30kg/10a, respectively. The cold-pretreatment for anthers was only treated to three nitrogen levels for 10 days at 10℃. The number of N-and S- pollen grains in vitro was compared to japonica type cultivars showing high sporophytic potential after 30 days in culture. Effect of day length on callus formation and on plant regeneration was different among cultivars. But S-pollen grains increased by the day length treatments in Seogwangbyeo and Pungsanbyeo. The rates of S-pollen grains and callus formation were significantly increased in 0 and 30kg/10a nitrogen levels. Callus formation and plant regeneration were remarkably increased by the interaction between nitrogen stress and cold pretreatment. Samgangbyeo, grown under nitrogen-free condition, showed the highest reaction as 37.4% and 12.0%, respectively. In vitro the development of N pollen grains form inoculated anther of tongil type cultivars was more noticeable than in japonica type cultivars, Tongil type cultivars seemed to be low callus formation because they have high gametophytic potential in vitro.
