Culture media for germination of excised embryos from grass seeds and in vitro culture of ovules from intergeneric crossing

The excised embryo cuture was examined to find the optimum condition for immature ovule culture in vitro, and the ovule transfer to culture media was conducted to raise the intergeneric hybrids between Festuca and Lolium genus. Excised embryos from Lolium multiflorum seeds were cultured in MS, N6, White’s media with various plant growth regulators to determine optimum culture conditions for embryo germination and growth. MS medium was the highest in germination rate, which was about same to germination rate of normal seeds. But the seedling growth of MS medium was poor compared to normal seeds. Gibberellin and kinetin were effective on germination of excised embryo, while BA and 2, 4-D were produced various abnormal seedlings including albino and malformed. After artificial pollination in vivo were conducted between Festuca arundinacea and Lolium multiflorum, the maturing ovules were transfered periodically into culture media to secure intergeneric hybrid seedlings. Seven days old ovules could germinate after 60 day’s culturing in medium. The 10~14 days old ovules were resulted over 60% in germination and produced healthy hybrid seedling. We concluded the optimum stage for ovule transfer was 10 to 14 days after pollination.
