Cation and proline content in F₃ seed-derived callus grown on the medium with and without NaCl were analyzed to determine reliable selection marker and to know the linkage analysis for salt tolerance in in vitro culture of rice. The results obtained are summarized as follows:Proline accumlation ability of linkage testers was statistically significantly different under salt stress, but cation content in the callus grown the medium with NaCl was not significantly different. The segregation ratio of proline content, which was divided into two groups based on the critical value of 300 ppm, was 9:7(low:high) in the crosses between 3 linkage testers(Ⅰ, Ⅹ, XI) and T1668. The genes related to proline accumulation were independent of Cl and wx of linkage group Ⅰ, bl-1 of linkage group Ⅹ. and be of linkage group XI. Variation in the F₂ population was even greater, and the range extended beyond that of both parents. The results of the cross implied that the ability to accumulate proline under salt stress is likely to be a quantitative character.
