A modified DNA-DNA in situ hybridization procedure was used to localize rRNA genes on chromosomes using cytological preparations. ³H-labeled 18S, 5.8S, and 26S maize rRNA genes specifically hybridized to the nucleolus organizor region(NOR) of pachytene and diplotene chromosme, indicating that 18S, 5.8S, and 26S rRNA genes were present in the NOR of maize chromosome 6. This also suggested that the in situ hybridization procedure used in this study could be applicable to localize other genes on chromosomes and investigate their structural characteristics using cytological preparations.
