Effects of subculture and cold storage on essential oil content and its composition in the callus of mentha viridis L.

Essential oil and menthol content were gradually decreased with passages of subculture. However, essential oil and menthol content were not changed even after 150 days when stored at low temperatue of 5 and 10℃. Cell growth at 5℃ was a little bit inhibited but browning rate of callus was much slower than at 10℃ after 150 days storage. Stored at 5℃ for 100 days long was appeared to be more effective way for both maintaining the ability of methol synthesis in the callus cells and essential oil content in avoiding of serial subculture with 25 days interval.
