Callus induction and culture for indigo production in polygonum tinctorium lour

This experiment was carried out to obtain basic information for in vitro production of indigo via callus cultures in Polygonum tinctorium. The results indicated that callus was induced well from leaf disc on B5 medium containing 1㎎/ℓ of 2,4-D. Callus grown well on B5 medium which containing 19g/ℓ of yeast extract and 2㎎/ℓ of 2,4-D under dark condition. Sucrose(2%) as carbon source and nitrate nitrogen as nitrogen source were good for callus growth. When phosphate was added twice much to the basic medium, callus growth was increased. Indigo synthesis in callus cells was confirmed through wave length scanning and TLC analysis.
