도라지의 잎과 줄기배양에 의한 體細胞胚 發生과 植物體 再分化

Somatic embryogenesis and plant regeneration from leaf and stem culture of Platycodon grandiflorum

These experiments were carried out to examine the effect of explant sources and plant growth regulators on somatic embryogenesis and plant regeneration. Leaf blade and stem explant of Platycodon grandiflorum were cultured on Murashige and Skoog(MS) medium supplemented with different concentrations of auxins and cytokinins. Leaf blade and stem explant were taken from the aseptically germinated young plants in vitro. Leaf blade and stem explant cultured on MS medium supplemented with 2,4-D and kinetin were observed to form compact callus_ The somatic embryos appeared to be initiated on the callus surface in the initial culture medium after 50 days of culture. Somatic embrvogenesis from leaf blade was the most effective on medium with 1.0 ㎎/L 2,4-D and 1.0 ㎎/L kinetin, that from stem was on medium with 0.5 ㎎/L 2,4-D and 0.5 ㎎/L kinetin. In combination of NAA and BA, somatic embryogenesis was observed on medium with 0.5 ㎎/L NAA and 1.0 ㎎/L BA in leaf blade and stem explant. Leaf blade tissue was directly formed shoots from explant surface without callus induction on medium with 0.5 ㎎/L NAA and 0.5 ㎎/L BA after 7 days of culture. When the somatic embryos were subcultured on 0.5 ㎎/L NAA and 1.0 ㎎/L BA, plantlets regenerated from somatic embryos were having normal shoots and roots.
