Direct somatic embryogenesis and plant regeneration from root suspension culture of Bupleurum falcatum L.

To improve an efficiency of in vitro propagation of Bupleurum falcatum L., direct somatic embryogenesis and plant regeneration have been attempted in suspension culture of roots differentiated from leaf-derived callus. Rooting from callus suspension culture was better in high concentration than in low concentration of sucrose. Eighteen somatic embryos per 1cm long root segment were induced in MS liquid medium containing 0.5㎎/ℓ 2,4-D. As the concentration of 2,4-D increased, the frequency of somatic embryos at globular stage was higher. A number of somatic embryos at cotyledonary stage were observed in the media supplemented with 0.1-0.5㎎/ℓ 2,4-D. Somatic embryos in the aggregates (somatic embryo clumps) were most frequently found in the medium containing 0.1㎎/ℓ 2,4-D, of which half were consist of 6 to 10 somatic embryos. The frequency of plant regeneration from somatic embryos was highest in 1/4 strength MS medium:11.7 % and 62.5% respectively from somatic embryos at torpedo and cotyledonary stages.
