Factors affecting cell growth in suspension culture of polygonum tinctorium lour

This experiment was carried out to analyze the factors affecting callus growth in suspension culture of Polygonum tinctorium for indigo and indirubin production. The B5 liquid medium was good for growth of callus. Sucrose was more effective than glucose as carbon sources and nitrate was better than ammonium as nitrogen source. Increased concentration of phosphate had little effect on callus growth and 2,4-D(2㎎/ℓ) alone was enough for callus growth. Optimum RPM was 130 and light condition was better than dark for callus growth. Modified B5 liquid medium without ammonium nitrogen but with yeast extract 1g/ℓ increased 1.2 times callus growth compared with original B5.
