멧대추(山棗)의 體細胞胚 發生에 의한 植物體 再分化

Plant regeneration through somatic embryogenesis of Zizyphus jujuba M.

Plant regeneration through somatic embryogenesis was conducted by culturing mature zygotic embryos of Chinese jujube(Zizyphus jujuba M.). About 20% of the calli obtained on MS medium supplemented with 0.1 ㎎/ℓ BA and 1.0 ㎎/ℓ 2,4-D was embryogenic. The embryogenic calli were transferred onto MS medium supplemented with different concentrations of BA, 2-iP or zeatin for somatic embryo formation. Normal somatic embryos showing different developmental stages could be induced on BA containing media. However, many abnormal somatic embryos were observed on the media containing either 2-iP or zeatin. Both somatic embryo germination and plant regeneration were readily achieved on MS medium containing 0.2 ㎎/ℓ GA₃. However the treatment of ABA, ABA plus GA₃ or activated charcoal resulted in poor response. Air-drying treatment for somatic embryo germination Was not effective. Some positive and negative aspects concerning somatic embryogenesis of Zizyphus jujuba were also discussed.
